Q1. Can RNA samples be directly used for library preparation with the LeXPrep kit?

A: No. This kit is compatible only with gDNA or cDNA. For RNA samples, reverse transcription to generate cDNA is required prior to starting the library preparation protocol.

Q2. What is the recommended input range, and how should I handle inputs exceeding this range?

A: The LeXPrep kit supports an input range of 50–2,000 ng of gDNA or cDNA. If the input amount exceeds 2,000 ng, we recommend splitting the sample into multiple reactions to maintain optimal amplification efficiency.

Q3. What is the expected insert size, and how should I select the sequencing read length?

A: The primary peak of PCR products generated by the LeXPrep kit is approximately 270 bp. We recommend PE150 (paired-end 150 bp) sequencing to ensure high-quality data coverage.

Q4. What is the recommended sequencing data volume for IG/TR immune repertoire analysis?

A: A minimum of 0.3 Gb of data is recommended to detect clones at 0.01% abundance with a 200 ng input. To enhance sensitivity for lower-frequency clones, the data volume should be increased accordingly.

Q5. Is the LeXPrep kit suitable for Minimal Residual Disease (MRD) monitoring?

A: Yes. The kit includes IG and TR Primer Mixes provided in separate tubes, allowing for flexible combination in a single amplification reaction. Due to its high sensitivity and ability to detect low-frequency variants, LeXPrep is an ideal solution for MRD monitoring technology development and clinical research applications.
